Why model Timp2

Distinctive among the family, Timp2 is the mouse ortholog of human TIMP2 and encodes the only member that efficiently blocks MT1-MMP (Mmp14) while paradoxically being required for pro-MMP2 activation, because its C-terminal domain tethers pro-MMP2 to MT1-MMP on the cell surface so that a neighboring uninhibited MT1-MMP molecule performs the activating cleavage. The N-terminal domain provides canonical zinc chelation through its Cys1 start, and TIMP2 additionally signals through integrin alpha 3 beta 1 and SHP-1 to suppress VEGFR2 phosphorylation, giving MMP-independent antiangiogenic activity. Nulls are viable with loss of MMP2 activation and altered angiogenesis, so germline deletion is informative but conflates inhibitory and activating roles, which argues for C-terminal or Cys1 knockins that separate them plus conditional alleles for stromal versus endothelial sources. Timp3 and Timp4 partly overlap on MT1-MMP, so plan compound genetics and align alleles with collagen invasion, corneal neovascularization, or MMP2 zymography endpoints.
