As the mouse ortholog of human TRADD, Tradd encodes the TNFR1-associated death domain protein, the first adaptor recruited to the ligated TNFR1 death domain, using its own C-terminal death domain for receptor binding and an N-terminal TRAF-interacting domain to dock TRAF2, thereby delivering cIAP1 and cIAP2 and enabling the ubiquitin-dependent assembly that drives NF-kB and MAPK activation. In the alternative configuration TRADD nucleates cytosolic complex II with FADD and caspase-8 to execute apoptosis, and it also participates in DR3 and TLR-driven signaling. Nulls are viable and fertile, showing reduced TNF-induced NF-kB and marked resistance to TNF-dependent apoptosis while retaining RIPK1-dependent necroptosis, so germline deletion is an efficient epistasis tool and conditional alleles resolve hepatocyte or intestinal epithelial requirement. Because RIPK1 can be recruited independently of TRADD, compound Ripk1 genetics clarifies the split, and alleles should follow hepatotoxicity, colitis, or death pathway endpoints.
