Why model Uqcrc1

Uqcrc1 is the mouse ortholog of human UQCRC1 and encodes core protein 1 of complex III, a large matrix-facing subunit that, with core protein 2, adopts a mitochondrial processing peptidase-like fold and forms the structural platform onto which cytochrome b, cytochrome c1 and the Rieske protein assemble in the dimeric bc1 enzyme. These core subunits carry no redox center but are required for holoenzyme stability and for processing events during maturation of the Rieske subunit presequence, so their reduction destabilizes the entire complex and lowers ubiquinol to cytochrome c oxidoreductase flux. Human variants have been linked to complex III deficiency and, separately, reported in familial parkinsonism with dopaminergic vulnerability, making both a conditional null with dopaminergic Cre drivers and a patient-matched missense knockin defensible depending on whether bioenergetic failure or neurodegeneration is the question.
