Uqcrq, ortholog of human UQCRQ, encodes the small ubiquinone-binding protein subunit of complex III that packs against cytochrome b and helps shape the environment of the quinone sites where the bifurcated Q cycle operates, moving electrons from ubiquinol to the Rieske iron-sulfur protein and to the b hemes. A homozygous missense substitution in this subunit was identified as a cause of severe complex III deficiency with Leigh-like encephalopathy and profound psychomotor impairment, establishing that even a small structural subunit can be rate limiting for holoenzyme function. That makes a patient-matched knockin the most faithful design, since it preserves partial assembly and reproduces graded respiratory chain failure, whereas a floxed conditional null answers the separate question of where complex III abundance constrains oxidative metabolism.
