Vegfa, the mouse ortholog of human VEGFA, encodes the dimeric heparin-binding growth factor spliced into 120, 164, and 188 residue isoforms that differ in exon 6 and 7 matrix retention and signal through KDR (VEGFR2) autophosphorylation to PLCG1, PKC, and ERK, with FLT1 acting as ligand sink and neuropilins as coreceptors during DLL4-Notch-governed tip cell selection. Unusually, this locus is haploinsufficient: even heterozygous deletion is lethal around E11 from failed endothelial assembly, so conditional floxed alleles paired with tightly validated Cdh5, Sox2, or lineage-restricted Cre drivers are mandatory rather than optional, and isoform-restricted knockins expressing only the 120 or 188 form dissect gradient formation from bulk mitogenic dose. Humanization matters because bevacizumab and ranibizumab do not neutralize mouse VEGF-A. Vegfb and Pgf engage FLT1 alone and cannot restore KDR signaling, so map alleles onto oxygen-induced retinopathy, permeability, or tumor angiogenesis endpoints.
