Gabrb1 encodes the β1 GABA-A receptor subunit, ortholog of human GABRB1, one of three β isoforms that contribute the principal side of the orthosteric GABA binding site at the β and α interface and that carry the transmembrane residues determining sensitivity to etomidate, propofol and loreclezole. Relative to β2 and β3 it is the least abundant, with expression concentrated in hippocampus, septum and specific hypothalamic and brainstem nuclei, and it is the sole β subunit in the imprinted-adjacent chromosome 4p12 cluster with GABRA2, GABRA4 and GABRG1. Because β2 and β3 substitute broadly during assembly, a single Gabrb1 deletion is frequently silent; plan compound conditional alleles, quantify pentamer composition biochemically, and read out anesthetic sensitivity alongside EEG.
