Gria3 is X-linked in both species and, as the ortholog of human GRIA3, encodes GluA3, an AMPA receptor subunit with a short cytoplasmic tail that supports constitutive, activity-independent receptor cycling through PICK1 and GRIP interactions, in contrast with the regulated insertion driven by GluA1. Human GRIA3 missense variants produce X-linked intellectual disability with both loss-of-function and gain-of-function gating changes, including slowed deactivation and shifted agonist potency, which is why patient-specific knockin alleles are usually more informative than deletion. Constitutive nulls are viable with subtle synaptic phenotypes, so conditional alleles are reserved for cell-type attribution rather than for bypassing lethality. Because Gria1 and Gria2 readily substitute in heteromers, score subunit-resolved currents and rectification, and use humanized alleles when testing modulators with species-divergent binding sites.
