Why model Grin1
Grin1, the mouse ortholog of human GRIN1, encodes GluN1, the obligatory subunit present in every NMDA receptor tetramer, contributing the glycine and D-serine binding S1S2 clamshell that must be co-occupied with glutamate on GluN2 subunits for channel opening. Eight GluN1 splice variants generated by cassette exon 5 and the C1, C2, and C2 prime tails control proton sensitivity, PKC modulation, and PDZ-dependent trafficking, while the M2 asparagine sets magnesium block and calcium permeability.
