Why model Grin2c

Grin2c, the mouse ortholog of human GRIN2C, encodes GluN2C, an NMDA receptor subunit whose expression is concentrated in cerebellar granule cells, thalamic relay nuclei, oligodendrocyte lineage cells, and some astrocytes, appearing after the second postnatal week. GluN2C-containing receptors are biophysically distinctive: low open probability, weak magnesium block relative to GluN2A and GluN2B, low single-channel conductance, and slow deactivation, which lets them pass current near resting potential and act as tonic rather than coincidence-detecting conductances. Nulls are viable with mild cerebellar motor learning deficits, so conditional alleles serve cell-type attribution in thalamocortical or glial compartments rather than lethality rescue. Because GluN2A and GluN2B dominate most forebrain synapses and can populate triheteromeric receptors with GluN2C, interpret loss-of-function data with subunit-resolved pharmacology. Positive allosteric modulators selective for GluN2C and GluN2D have species-divergent sites, making humanized ligand-binding or amino-terminal domain knockins the right choice for translational pharmacology endpoints.
