The inhibin beta A subunit is encoded by Inhba, the mouse ortholog of human INHBA, which supplies the polypeptide that homodimerizes into activin A, heterodimerizes into activin AB, and pairs with the alpha subunit to form inhibin A. Activin A signals through ActRIIA or ActRIIB with ALK4 to phosphorylate SMAD2 and SMAD3, is neutralized extracellularly by follistatin, and drives mesoderm induction, pituitary FSH synthesis, wound repair, heterotopic ossification, and tumor-associated cachexia. Nulls die shortly after birth with cleft palate, absent whiskers, and incisor defects, so conditional deletion is necessary for adult muscle, immune, and cancer endpoints. Subunit-swap knockins, in which Inhbb coding sequence replaces Inhba, remain the cleanest test of whether phenotypes reflect ligand identity or expression domain, and humanization enables selectivity testing of anti-activin A antibodies. Because Inhbb partially substitutes through shared receptors, plan compound conditional genetics before attributing a phenotype to activin A alone.
