Encoding pyruvate dehydrogenase kinase isozyme 2, Pdk2 is the mouse ortholog of human PDK2 and phosphorylates serine residues on the E1 alpha subunit to switch off the pyruvate dehydrogenase complex, acting as a direct brake on glycolytic carbon entry into the TCA cycle. This isoform is the most broadly expressed of the four, the most sensitive to inhibition by pyruvate and by dichloroacetate, and is stimulated by elevated acetyl-CoA to CoA and NADH to NAD ratios, so it enforces the classic fatty acid to glucose oxidation switch in heart, muscle and liver. Design choices split cleanly: a conditional null asks where tonic PDH suppression is required and typically increases glucose oxidation, while humanization enables testing of dichloroacetate analogs and newer inhibitors against the human pharmacophore. Real redundancy exists across Pdk1, Pdk3 and Pdk4, so plan compound conditional genetics before interpreting a single knockout.
