Nomenclature deserves care here: Pdk1 and human PDK1 denote pyruvate dehydrogenase kinase isozyme 1, a mitochondrial matrix kinase distinct from the PI3K effector Pdpk1. The enzyme phosphorylates serine residues on the E1 alpha subunit of the pyruvate dehydrogenase complex, principally site 1, inactivating the complex and blocking pyruvate entry into the TCA cycle, thereby diverting carbon toward lactate. It is allosterically inhibited by pyruvate and by dichloroacetate and induced transcriptionally by HIF1 alpha, placing it at the switch that installs aerobic glycolysis in hypoxic and tumor tissue. Conditional nulls crossed to cardiomyocyte or tumor-restricted Cre drivers test whether relieving pyruvate dehydrogenase inhibition reverses the glycolytic phenotype, while kinase-dead or hypoxia-response-element mutant knockins isolate catalysis from induction. Because Pdk2, Pdk3 and Pdk4 phosphorylate overlapping sites with different tissue distributions and strong fasting induction, single-isoform deletion frequently yields a masked metabolic phenotype and compound alleles should be planned early.
