Why model Sost
Sost is the mouse ortholog of human SOST and encodes sclerostin, an osteocyte-derived, secreted cysteine-knot protein of the DAN family that binds the first beta-propeller of LRP5 and LRP6, using LRP4 as a presenting platform, to block Wnt, Frizzled, and LRP coreceptor assembly and suppress beta-catenin signaling in osteoblasts, with a secondary reported BMP-antagonist activity. Mechanical loading downregulates it, and human loss of function causes sclerosteosis or, through deletion of the ECR5 enhancer roughly 35 kilobases downstream, van Buchem disease, both high bone mass conditions that validated antibody blockade. This makes the allele fork unusually crisp: osteocyte-restricted conditional nulls using Dmp1-Cre test cell of origin and adult loading responses, ECR5 enhancer deletion models the human regulatory disease, and humanizing the LRP-binding loop enables direct antibody pharmacology. Sostdc1 shares the same LRP-binding mode, so consider compound alleles when dental or renal endpoints accompany skeletal readouts.
