Why model Aldoa
Aldoa encodes the mouse ortholog of human ALDOA, the class I fructose-bisphosphate aldolase that assembles into a homotetramer and cleaves fructose-1,6-bisphosphate into dihydroxyacetone phosphate and glyceraldehyde-3-phosphate through a Schiff base intermediate formed on an active site lysine. Aldolase A carries most glycolytic flux in skeletal muscle and erythrocytes, and it also binds filamentous actin, WASP family proteins, and the V-ATPase, so loss perturbs cytoskeletal and vesicular biology alongside ATP supply. Recessive human variants produce nonspherocytic hemolytic anemia with rhabdomyolysis, which argues for point knockins rather than deletion when disease fidelity is the goal. Because Aldob and Aldoc are catalytically competent paralogs with overlapping tissue windows, single locus deletion can be buffered wherever they are co-expressed, so compound alleles belong in the plan early.
