Why model Dlg2
Encoding PSD-93, also called chapsyn-110, Dlg2 is the mouse ortholog of human DLG2 and a palmitoylated membrane-associated guanylate kinase whose N-terminal cysteines drive raft targeting and whose PDZ1 and PDZ2 domains bind the ESDV motif of GluN2A and GluN2B, clustering NMDA receptors and Kv1 channels at the postsynaptic density while its guanylate kinase domain recruits GKAP and the Shank and Homer scaffold layer. It preferentially stabilizes GluN2A-containing receptors in mature synapses, giving it a developmental profile distinct from SAP102. Rare deletions and exonic copy number variants at the human locus are established risk factors for schizophrenia and intellectual disability, making dosage-sensitive alleles biologically relevant. Constitutive nulls are viable with mild synaptic phenotypes, so a heterozygous or conditional deletion better mirrors human haploinsufficiency, and Camk2a-Cre or interneuron drivers assign circuit contributions. Dlg1, Dlg3 and Dlg4 compensate substantially, so quantify all four paralogs after deletion.
