Dlg3 encodes SAP102, ortholog of human DLG3, a membrane-associated guanylate kinase distinguished from PSD-95 by the absence of N-terminal palmitoylation and by an alternatively spliced insert that binds mPins and exportin, features that make it the dominant NMDA receptor scaffold in immature synapses where it preferentially associates with GluN2B-containing receptors and supports their delivery before PSD-95 and PSD-93 take over. Its PDZ domains also engage GluA2 and the exocyst, giving it a role in receptor trafficking as well as anchoring. Truncating mutations in the X-linked human gene cause nonsyndromic intellectual disability, so hemizygous male mice provide a directly comparable dosage model. Constitutive nulls are viable with impaired learning and altered synaptic plasticity, though the developmental window matters: phenotypes are strongest early and are progressively masked as Dlg4 and Dlg2 expression rises.
