PSD-95, encoded by Dlg4 as the ortholog of human DLG4, is the dominant scaffold of the excitatory postsynaptic density, anchored by palmitoylation of Cys3 and Cys5 and organizing receptors through PDZ1 and PDZ2 binding to the GluN2 ESDV motif and to transmembrane AMPA receptor regulatory protein C-termini including γ-8, while PDZ2 recruits nNOS and the guanylate kinase domain assembles the GKAP, Shank and Homer layers. Its abundance sets AMPA receptor content per synapse, and disruption of the nNOS interaction is an established stroke neuroprotection strategy. Constitutive nulls are viable but show markedly enhanced long-term potentiation, absent long-term depression and impaired spatial learning, so lethality is not a constraint; a conditional null with Camk2a-Cre isolates forebrain contributions and a palmitoylation-site or PDZ-binding knockin separates membrane anchoring from protein interaction. Because Dlg1, Dlg2 and Dlg3 upregulate and partly substitute, compound conditional genetics and quantitative postsynaptic density proteomics should be planned from the start.
