De novo GRIN2D variants such as V667I cause severe developmental and epileptic encephalopathy through gain of function, prolonging current and increasing agonist potency, so knockin alleles model the disease far better than deletion, which yields viable mice with modest behavioral change. Grin2c overlaps functionally in some populations and GluN2A or GluN2B substitutes elsewhere, so combine subunit-resolved recordings with interneuron-restricted Cre drivers and reserve humanized alleles for GluN2D-preferring modulator testing.
