Why model Il13
Il13 corresponds to human IL13 and encodes a type 2 cytokine that signals through the type II receptor composed of IL-4Ralpha and IL-13Ralpha1 to activate STAT6, with IL-13Ralpha2 acting as a high-affinity decoy, driving goblet cell metaplasia, smooth muscle hyperresponsiveness, periostin production, and fibroblast activation in airway and skin disease. It sits within the type 2 locus immediately adjacent to Il4 and near Il5 under shared conserved noncoding elements including Rad50 region control sequences, so targeting cassettes and floxed designs must be checked for cis effects on neighboring genes, a frequent source of misattributed phenotypes. Humanization is the correct allele for dupilumab, tralokinumab, and lebrikizumab class pharmacology, while reporter knockins localize the producing Th2, ILC2, or mast cell source.
