Why model Il12b
The p40 subunit encoded by Il12b, mouse ortholog of human IL12B, is the shared chain of two distinct heterodimeric cytokines, interleukin 12 when paired with p35 and interleukin 23 when paired with p19. Interleukin 12 drives STAT4 dependent Th1 differentiation and interferon gamma, whereas interleukin 23 sustains RORgt dependent Th17 and ILC3 programs with interleukin 17 and interleukin 22 output, so a single Il12b null collapses both axes at once and cannot distinguish them. This is precisely the ambiguity that made p40 blockade with ustekinumab clinically effective yet mechanistically opaque, and why selective p19 blockade later outperformed it in psoriasis. Rational design pairs Il12b alleles with Il12a and Il23a single nulls to partition the two cytokines, uses conditional deletion in dendritic cells or macrophages to identify the producing compartment, and adopts humanization when p40 antibody pharmacology rather than pathway ablation is the endpoint. Homodimeric p40 can also antagonize receptor engagement, complicating partial loss interpretation.
