Il12a, the mouse ortholog of human IL12A, encodes the p35 subunit, and its central design trap is subunit sharing: p35 heterodimerizes with p40 from Il12b to form bioactive interleukin 12, and with EBI3 to form interleukin 35, an inhibitory cytokine of regulatory T and B cells. Interleukin 12 acts through IL-12Rbeta1 and IL-12Rbeta2 to trigger JAK2 and STAT4, imprinting Th1 commitment and interferon gamma production, while interleukin 35 signals through gp130 with IL-12Rbeta2 to activate STAT1 and STAT4 in a suppressive configuration. A germline Il12a null therefore removes a proinflammatory and an anti-inflammatory cytokine simultaneously, so phenotypes read as net effects rather than clean interleukin 12 loss. Dissection requires triangulation across Il12b, Ebi3, and Il12rb2 alleles, plus conditional Il12a deletion with dendritic cell or B cell restricted Cre drivers, and humanization when the endpoint is ustekinumab-class or p35 directed antibody pharmacology.
