Why model Il17a
Il17a is the mouse ortholog of human IL17A and encodes a cystine-knot cytokine acting as a homodimer, and also as a heterodimer with interleukin 17F, on the IL-17RA and IL-17RC receptor complex, recruiting the adaptor Act1 encoded by Traf3ip2 plus TRAF6 to activate NF-kB, MAPK signaling, and transcript stabilization of CXCL1, CXCL2, CSF3, and antimicrobial peptides in epithelium and stroma. Producers are RORgt dependent Th17 cells, gamma delta T17 cells, ILC3s, and MAIT cells, and human IL17RA or IL17F lesions cause chronic mucocutaneous candidiasis while excess drives psoriasis, the indication where secukinumab and ixekizumab succeeded. Because Il17a and Il17f are adjacent and coregulated, single nulls give partial phenotypes and heterodimer contributions are missed, so compound alleles or receptor deletion are frequently required. Fate mapping alleles such as Il17a-Cre distinguish current from historical expression, and conditional deletion in gamma delta versus CD4 compartments is what actually assigns the pathogenic source.
