Why model Il18
Il18 encodes interleukin 18, the mouse ortholog of human IL18 and an interleukin 1 family member synthesized as an inactive precursor constitutively stockpiled in epithelium, keratinocytes, and macrophages, then cleaved by caspase-1 downstream of NLRP3 or NLRC4 inflammasomes, or by caspase-8, neutrophil proteases, and granzyme B, before release through gasdermin D pores. Mature cytokine engages IL-18R1 with the accessory chain IL18RAP to recruit MyD88 and IRAK4, driving NF-kB and, together with interleukin 12, potent interferon gamma output from Th1, NK, and ILC1 cells, while the secreted decoy interleukin 18 binding protein sets the free ligand concentration and rises in macrophage activation syndrome. Allele design should follow that regulation: a conditional null tests requirement, an uncleavable precursor knockin isolates processing, and humanization supports both neutralizing antibody and decoy-resistant agonist programs. Because Il18 and Il1b share upstream inflammasome machinery, attribute phenotypes with ligand-specific alleles rather than caspase-1 deletion.
