Il1a, mouse ortholog of human IL1A, encodes interleukin 1 alpha, a cytokine whose full-length precursor is already bioactive, carries a nuclear localization sequence, tethers to the plasma membrane, and is released as an alarmin when cells die by necrosis or necroptosis, with calpain dependent processing enhancing potency. This makes its regulation fundamentally different from interleukin 1 beta, which requires inflammasome caspase-1 cleavage, even though both signal through IL-1R1 with IL1RAP to recruit MyD88, IRAK4, and IRAK1 and activate NF-kB and p38. Interleukin 1 alpha therefore dominates sterile injury, ultraviolet damage, and tumor associated inflammation, and its nuclear pool contributes transcriptionally in ways a secretion-focused readout misses. Distinguish these functions with a conditional null, a nuclear localization deficient or membrane-anchored knockin, and humanization for bermekimab-class antibodies. Because Il1b and Il1rn act on the same receptor, compound alleles or Il1r1 deletion are needed to bound total interleukin 1 activity in any model.
