Why model Il2
Il2 is the mouse ortholog of human IL2 and encodes a four alpha helix bundle cytokine that assembles a trimeric receptor of IL-2Ralpha, IL-2Rbeta, and the common gamma chain, activating JAK1 and JAK3 with STAT5 to control regulatory T cell fitness, effector CD8 expansion, and activation induced cell death. Because high-affinity capture depends on constitutive CD25 on regulatory T cells, interleukin 2 availability is a tolerance checkpoint, and germline Il2 or Il2ra nulls develop lethal lymphoproliferation with autoimmune hemolytic anemia and colitis rather than immunodeficiency, a phenotype that constrains any breeding scheme. Allele class should follow the endpoint: conditional deletion in CD4 T cells asks who supplies the cytokine, knockins encoding not-alpha or CD25-biased muteins model low dose interleukin 2 therapy, and humanization supports aldesleukin and engineered agonist pharmacology.
