Why model Il27
Il27 encodes the p28 subunit, mouse ortholog of human IL27, which pairs with EBI3 to form interleukin 27 and signals through IL-27Ralpha, also called WSX-1, together with gp130 to activate both STAT1 and STAT3. That dual output explains its split personality, with early amplification of Th1 commitment and T-bet alongside potent suppression of Th17 differentiation, induction of interleukin 10 producing Tr1 cells, and upregulation of inhibitory receptors including PD-L1 and TIM-3 on T cells. Two design subtleties matter. EBI3 is shared with interleukin 35, so an Ebi3 null removes two cytokines while an Il27 null removes only p28 containing species, and unpaired p28 can antagonize gp130 signaling, meaning overexpression and deletion are not simple mirror images. Conditional deletion in dendritic cells or macrophages assigns the source, Il27ra alleles test receptor requirement in T cells, and humanization supports antibody pharmacology where the immunosuppressive arm is the therapeutic target.
