Roughly half of nulls die in utero from yolk sac vasculogenesis failure and survivors succumb to multiorgan autoimmune inflammation within weeks unless crossed onto immunodeficient backgrounds, while human loss of function causes very early onset inflammatory bowel disease and latency peptide mutations cause Camurati-Engelmann disease. Immune-restricted conditional nulls, activation-site knockins, and humanized alleles for antibody testing answer distinct questions, with Tgfb2 and Tgfb3 sharing receptors.
