Tgfb1 is the mouse ortholog of human TGFB1 and encodes the prototypical isoform, secreted as a latent complex in which the mature disulfide-linked dimer is shielded noncovalently by its latency-associated peptide and tethered to LTBP1 or LTBP3 in matrix, or to GARP on regulatory T cells. Immune-restricted conditional nulls, activation-site knockins, and humanized alleles for antibody testing answer distinct questions, with Tgfb2 and Tgfb3 sharing receptors.
