Epithelial conditional deletion using K14-Cre separates palatal seam requirements from mesenchymal signaling, patient knockins address aortic and myocardial disease mechanism, and humanization supports isoform-selective pharmacology. Although Tgfb1 and Tgfb2 use the same receptor pair, their nonoverlapping null phenotypes show compensation is limited in vivo, so treat isoform identity as functionally meaningful.
