Why model Smad5
Smad5, the mouse ortholog of SMAD5, encodes a BMP receptor-regulated SMAD phosphorylated at its carboxy-terminal SSVS motif by ALK1, ALK2, ALK3 and ALK6, after which it assembles heterotrimers with SMAD4 and occupies GC-rich BMP response elements, with linker phosphorylation by GSK3 and MAPK recruiting SMURF ligases to terminate signaling. Germline deletion is embryonic lethal at midgestation with defective yolk sac and amnion angiogenesis, cardiac looping abnormalities and primordial germ cell deficits, so conditional alleles with endothelial or hepatocyte Cre drivers are necessary for postnatal analysis, particularly for hepcidin regulation and iron homeostasis where BMP6 and ALK3 input converge. Redundancy here is genuine and quantitative: Smad1 and Smad9 use the same phosphoacceptor motif and the same response elements, and compound Smad1 and Smad5 deletion produces far more severe phenotypes, so plan double conditional genetics before attributing a BMP phenotype to this locus alone.
