Every branch of the TGF beta superfamily converges on Smad4 (ortholog of human SMAD4), the sole common mediator SMAD, which trimerizes through its MH2 domain with two receptor-phosphorylated SMADs, either SMAD2 and SMAD3 downstream of TGF beta and activin or SMAD1, SMAD5 and SMAD9 downstream of BMP, and supplies nuclear export control plus the transcriptional activation surface for the complex. Because it is shared, deletion collapses both branches at once, which is informative for tumor suppression but confounds branch attribution. Germline loss is early embryonic lethal at gastrulation, and somatic inactivation defines juvenile polyposis and is a canonical event in pancreatic ductal adenocarcinoma, where conditional nulls combined with Kras G12D and Trp53 alleles reproduce the human progression sequence. When the question is branch-specific, receptor-SMAD conditional alleles are preferable to Smad4 deletion, and MH2 interface knockins test complex assembly without abolishing protein expression.
