Smarca4 is the mouse ortholog of human SMARCA4 and encodes BRG1, one of two mutually exclusive ATPase motors of the BAF, PBAF, and non-canonical BAF chromatin remodeling complexes, using HSA, ATPase, and bromodomain modules together with SMARCB1, ARID1A, and DPF subunits to slide nucleosomes and maintain enhancer accessibility for lineage transcription factors. Inactivating mutations define small cell carcinoma of the ovary hypercalcemic type and thoracic SMARCA4-deficient undifferentiated tumor, and recur in lung adenocarcinoma, medulloblastoma, and Burkitt lymphoma, while distinct germline missense variants cause Coffin-Siris syndrome, so both null and hypomorphic allele classes are biologically legitimate. Germline deletion is lethal at the peri-implantation stage, making conditional nulls mandatory for tissue studies; ATPase-dead knockins separate remodeling from scaffolding, and humanized alleles serve variant classification. The paralog Smarca2 (BRM) substitutes in many contexts and underlies the well-documented synthetic lethal dependency of BRG1-deficient cells, so compound conditional design and inducible Cre timing are central to interpretation.
