Why model Cox17
Copper trafficking into cytochrome c oxidase begins with Cox17, the mouse ortholog of human COX17, a small twin CX9C intermembrane space metallochaperone that binds copper through conserved cysteines and hands it to SCO1 for the CuA site in MT-CO2 and to COX11 for the CuB site in MT-CO1. Loss of this single relay point compromises both metal centers simultaneously, which is why germline deletion in mouse arrests development near implantation, an established lethality that makes conditional alleles mandatory for any postnatal or tissue specific question. A floxed null therefore defines tissue tolerance for complex IV loss, while cysteine to serine knockins dissect metal binding from protein interaction. Cox19 and Cox23 participate in related redox and copper signaling but do not substitute for Cox17.
