Why model Cox5a
Cox5a encodes the mouse ortholog of human COX5A, a nuclear encoded matrix side subunit of cytochrome c oxidase that packs against MT-CO1 and contributes to allosteric regulation of the enzyme, including the relief of ATP mediated inhibition by 3,5-diiodothyronine binding described for this subunit. Subunit 5A is incorporated during assembly of the catalytic core, so its loss destabilizes the holoenzyme and produces isolated complex IV deficiency, and human recessive variants cause failure to thrive with lactic acidosis and pulmonary hypertension. Design therefore separates two questions: a conditional null tests absolute tissue requirement, while a missense knockin preserves assembly and interrogates regulation, which is the more informative allele if hormonal or nucleotide control is your target. No paralogous subunit replaces 5A. Use Ckmm-Cre, Myh6-MerCreMer, or Nes-Cre with complex IV spectrophotometry, respirometry across ATP to ADP ratios, echocardiography, and blue native assembly profiling.
