Why model Cox6c
Cox6c is the mouse ortholog of human COX6C and encodes a small nuclear encoded transmembrane subunit of cytochrome c oxidase added during the later assembly module together with subunits 7C and 8A, completing the peripheral shell around the mitochondrially encoded catalytic core. Subunits of this class stabilize the holoenzyme and its lipid interactions rather than participating directly in electron transfer, so depletion lowers assembled complex IV and cytochrome c oxidase activity while the heme and copper centers of surviving enzyme remain functional. The design decision is therefore whether you need absolute tissue requirement, addressed by a floxed conditional null, or a structural interface question, addressed by transmembrane substitutions that retain the subunit. No isoform paralog substitutes for 6C in somatic tissues. Direct deletion with Ckmm-Cre, Alb-Cre, or Rosa26-CreER, then quantify complex IV by spectrophotometry and histochemistry, respirometry, and assembly intermediates by blue native PAGE.
