Why model Cox6b1
Cox6b1, ortholog of human COX6B1, encodes a nuclear encoded subunit lying entirely on the intermembrane space face of cytochrome c oxidase, adopting a twin CX9C stabilized fold and sitting near the cytochrome c docking surface and the monomer to monomer interface of the dimeric enzyme. Because subunit 6B contributes to the electron entry region rather than to the catalytic metal centers, its loss reduces holoenzyme stability and turnover and yields isolated complex IV deficiency, and human recessive variants cause severe infantile encephalomyopathy. A conditional null therefore establishes tissue requirement, while patient missense knockins preserve assembly and better reproduce the partial activity relevant to human presentations. The testis enriched paralog Cox6b2 is not co-expressed in somatic tissues, so compensation should not be expected. Use Nes-Cre or Ckmm-Cre with cytochrome c oxidase activity, COX histochemistry, cytochrome c oxidation kinetics, and blue native monomer and dimer profiling.
