Why model Cox8a
Cox8a encodes the mouse ortholog of human COX8A, a small nuclear-encoded subunit of cytochrome c oxidase that packs against the mtDNA-encoded catalytic core (MT-CO1, MT-CO2, MT-CO3) and stabilizes the mature complex IV monomer during late assembly, supporting electron transfer from cytochrome c to oxygen and vectorial proton pumping. Because complex IV output is set by assembly efficiency rather than by transcript abundance of any accessory subunit, loss of Cox8a yields isolated complex IV deficiency with reduced respiratory reserve rather than global OXPHOS collapse. Allele choice follows the question: a conditional null with cardiac, skeletal muscle or neuronal Cre isolates cell-autonomous bioenergetic failure, whereas humanization supports testing of patient COX8A missense variants in situ. Note that Cox8b and Cox8c are expressed in restricted tissues, so confirm isoform usage before interpreting residual activity, and pair the allele with the endpoint, whether complex IV kinetics, BN-PAGE assembly intermediates, or organ stress physiology.
