Ndufa13, the mouse ortholog of human NDUFA13, encodes GRIM-19, a membrane arm subunit essential for complex I assembly that additionally functions as a negative regulator of STAT3, binding it and restraining transcriptional output, which gives the locus a dual identity spanning bioenergetics and growth signaling. Recessive human variants cause complex I deficiency with encephalopathy and hypertrophic cardiomyopathy, whereas reduced expression in thyroid and other tumors correlates with STAT3 activation. Germline deletion is not survivable, so conditional alleles are required, and the choice of Cre determines which identity is interrogated: cardiomyocyte or neuronal deletion for bioenergetic failure, epithelial or immune deletion for STAT3-dependent phenotypes. Separation-of-function knockins that disrupt the STAT3 interaction surface while preserving assembly competence are the only clean way to distinguish the two roles and are the design most often needed.
