Ndufs4, the mouse ortholog of human NDUFS4, encodes an accessory subunit required for final assembly and stabilization of the complex I N module, the flavin-containing NADH oxidation domain, and it is a substrate for PKA-dependent phosphorylation that tunes holoenzyme stability. Loss does not abolish the enzyme outright but leaves a stalled subcomplex lacking NDUFV1, NDUFV2 and NDUFS6, so NADH oxidation fails while membrane arm subunits persist. Constitutive nulls are viable at birth and then develop progressive ataxia, breathing abnormalities and brainstem and cerebellar lesions, giving one of the most faithful Leigh syndrome models available and a proven platform for interventions such as mTOR inhibition or chronic hypoxia. Conditional floxed alleles let the neuron versus glia versus muscle question be separated, since whole-body loss confounds cell-autonomous interpretation. Choose the conditional null with a defined neuronal Cre for lesion mapping, and reserve the germline null for survival and therapeutic response endpoints.
