Ndufs4, the mouse ortholog of human NDUFS4, encodes an accessory subunit required for final assembly and stabilization of the complex I N module, the flavin-containing NADH oxidation domain, and it is a substrate for PKA-dependent phosphorylation that tunes holoenzyme stability. Loss does not abolish the enzyme outright but leaves a stalled subcomplex lacking NDUFV1, NDUFV2 and NDUFS6, so NADH oxidation fails while membrane arm subunits persist.
