Tlr4 is the mouse ortholog of human TLR4 and encodes the lipopolysaccharide receptor, which requires the accessory protein MD-2 (Ly96) to form a pocket accommodating five of the six lipid A acyl chains while CD14 and LBP deliver ligand, and dimerization then triggers MyD88 and TIRAP signaling at the plasma membrane followed by TRAM and TRIF dependent IRF3 activation after endocytosis. Species specificity is a first-order design issue, since mouse and human MD-2 and TLR4 differ in their response to underacylated lipid A species, so lipid A mimetics, monophosphoryl lipid A adjuvants, and antagonists such as eritoran behave differently, which is why humanized Tlr4 and Ly96 knockins are used for adjuvant and sepsis pharmacology. The spontaneous P712H allele and complete nulls are both viable, so conditional deletion in myeloid, endothelial, or intestinal epithelial compartments is the refinement step, with endotoxin shock, neuropathic pain, or steatohepatitis endpoints dictating allele choice.
