Why model Gdf5
Known biochemically as cartilage-derived morphogenetic protein 1, Gdf5 is the mouse ortholog of human GDF5 and produces a BMP-family ligand with unusual selectivity for BMPR1B (ALK6) over ALK3, which it uses with BMPR2 or ActRIIA to drive SMAD1, SMAD5, and SMAD8 phosphorylation in the joint interzone, articular surface, tendon, and ligament. The spontaneous brachypodism allele is a Gdf5 null that shortens limb segments and eliminates specific joints, and human mutations span brachydactyly type C, Grebe and Hunter-Thompson chondrodysplasia, and gain-of-function symphalangism, with the common regulatory variant rs143383 in the R2 and R3 elements conferring osteoarthritis risk. Nulls are viable, so allele choice follows the question: conditional deletion for adult articular maintenance, coding knockins for chondrodysplasia mechanism, and enhancer knockins for the human osteoarthritis risk haplotype. Gdf6 and Gdf7 overlap in joint and tendon programs, so compound alleles and a joint-restricted Gdf5-Cre driver sharpen skeletal endpoints considerably.
