Why model Wnt16

Cortical bone is the compartment where Wnt16, the mouse ortholog of human WNT16, exerts its effect, since this osteoblast-derived Wnt signals through Frizzled with LRP5 or LRP6 and also engages noncanonical routes to suppress osteoclastogenesis both directly, by blunting RANKL-driven differentiation, and indirectly through osteoprotegerin induction. Human variation at the locus is among the strongest determinants of cortical bone mineral density and forearm fracture risk, and nulls are viable with thin, porous cortices and spontaneous fractures despite largely preserved trabecular bone, a compartment-selective phenotype that makes the gene valuable for skeletal fragility work. Osteoblast or osteocyte conditional deletion with Col1a1-Cre or Dmp1-Cre establishes cell of origin, gain-of-function knockins test whether restoring ligand suffices to thicken cortex, and humanization enables pathway agonist pharmacology. Redundancy is limited in the periosteal compartment, which is precisely why single-locus loss gives a clean readout, so cortical geometry and fracture endpoints remain directly interpretable.

What Wnt16 mouse models are available?

ingenious targeting laboratory offers 1 distinct Wnt16 (WNT16) catalog mouse model in the Wnt family, featuring conditional knockout and standard knockout. Researchers can order pre-developed catalog lines or request a custom mouse model, including humanized, knockin, and transgenic variations with verified germline transmission.
