Why model Wnt1
Wnt1 is the mouse ortholog of human WNT1 and encodes the founding Wnt ligand, palmitoleated on a conserved serine by Porcupine and exported through Wntless before engaging Frizzled receptors with LRP5 or LRP6 to inhibit the destruction complex and stabilize beta-catenin. Its critical developmental domain is the midbrain and hindbrain organizer, where it maintains an Engrailed, Pax2, and Fgf8 regulatory loop, and the spontaneous swaying allele plus targeted nulls lose midbrain and cerebellum with perinatal death.
