Wnt2 is the mouse ortholog of human WNT2 and encodes a canonical ligand of splanchnic mesenchyme that stabilizes beta-catenin through Frizzled with LRP6 in lung mesenchyme, where it specifies Nkx2-1 positive respiratory progenitors from foregut endoderm, and in placental and cardiac progenitor populations. Redundancy here is not hypothetical, since Wnt2b is coexpressed in the same ventral foregut mesenchyme and only compound Wnt2 and Wnt2b nulls abolish lung specification and produce lung agenesis.
