Why model Wnt2b

Wnt2b is the mouse ortholog of human WNT2B and encodes a canonical Wnt that stabilizes beta-catenin through Frizzled with LRP5 or LRP6 in ventral foregut mesenchyme, retinal ciliary margin progenitors, and the mesenchymal compartment supporting intestinal crypt stem cells. Biallelic human loss of function causes early onset chronic diarrhea with intestinal epithelial failure, establishing a non-redundant requirement in the human gut niche that a mouse single null does not obviously reproduce. Conditional deletion in intestinal subepithelial mesenchyme is therefore the informative design for niche questions, patient nonsense or missense knockins model the human enteropathy directly, and humanization supports agonist and R-spondin combination pharmacology. Wnt2 is coexpressed and functionally overlapping, most clearly in lung specification where only compound loss produces agenesis, so compound conditional alleles with a validated mesenchymal Cre driver belong in the initial plan whenever respiratory or intestinal niche output is the endpoint.
