Germline nulls die at gastrulation from failed mesodermal migration, so conditional floxed kinase domain alleles with lineage Cre are required. Fgfr2 and Fgfr3 overlap in ligand response and compensate in limb and skeleton, so compound alleles matter. Use isoform-restricted or point knockins when splice-specific or inhibitor questions drive the study.
