Why model Fgfr1

Fgfr1, mouse ortholog of human FGFR1, encodes a receptor tyrosine kinase with three immunoglobulin-like extracellular domains, an acid box, and a split intracellular kinase domain, requiring heparan sulfate proteoglycan as a ternary coreceptor for FGF binding and, for endocrine FGFs, klotho proteins. Alternative splicing of the second half of the third Ig domain generates IIIb and IIIc variants with divergent ligand specificity, and activated receptor recruits FRS2 alpha and PLC gamma to drive RAS-MAPK, PI3K and PKC output. Loss of function alleles cause Kallmann syndrome and hypogonadotropic hypogonadism, while activating P252R causes Pfeiffer syndrome and 8p11 fusions such as ZMYM2-FGFR1 drive myeloid neoplasms. Germline nulls die at gastrulation from failed mesodermal migration, so conditional floxed kinase domain alleles with lineage Cre are required. Fgfr2 and Fgfr3 overlap in ligand response and compensate in limb and skeleton, so compound alleles matter. Use isoform-restricted or point knockins when splice-specific or inhibitor questions drive the study.
