Why model Fgf21
Fgf21, the mouse ortholog of human FGF21, encodes an endocrine FGF whose weak heparan sulfate affinity permits circulation and whose activity requires the coreceptor beta-Klotho paired with FGFR1c on adipocytes, or FGFR1c in hypothalamus and hindbrain. Hepatic transcription is induced by PPARalpha during fasting and ketogenesis and by ATF4 during amino acid restriction and integrated stress response, and the protein raises adipose glucose uptake and adiponectin, promotes energy expenditure, and centrally suppresses sweet and alcohol preference while stimulating water intake.
